let-7e甲基化沉默在卵巢癌中的作用Epigenetic silencing of let-7e in ovarian cancer

摘要(Abstract):
目的:研究let-7e甲基化沉默在卵巢肿瘤中的作用。方法:采用q MSP、RTPCR法检测30例卵巢组织、A2780细胞系(A2780、A2780/CP)中let-7e甲基化水平及let-7e表达。采用DNA甲基化酶抑制剂5-氮杂-2'-脱氧胞苷(5-Aza-CdR)处理A2780/CP细胞,检测let-7e表达。结果:let-7e所在区域富含Cp G岛。高甲基化时,卵巢癌组织和细胞中let-7e表达量降低,let-7e表达与DNA甲基化呈负相关(r=-0.6194,P=0.0003)。5-Aza-CdR处理卵巢癌细胞后,let-7e表达明显升高。结论:卵巢癌组织和细胞中let-7e表达受DNA甲基化负调控。DNA甲基化下调let-7e表达可能参与卵巢癌的发生及进展,也与卵巢癌化疗耐药有关。
