下调microRNA-107表达对人子宫内膜样腺癌Ishikawa细胞增殖迁移和侵袭的作用Effects of abrogating expression of miR-107 on proliferation migration and invasion of endometrial carcinoma Ishikawa cells

摘要(Abstract):
目的:探讨下调microRNA-107(miR-107)表达对人子宫内膜样腺癌细胞系Ishikawa增殖迁移和侵袭能力的影响及可能机制。方法:将miR-107抑制剂(miR-107 inhibitor)瞬时转染Ishikawa细胞,下调miR-107表达。实时荧光定量PCR法(qRT-PCR)检测miR-107表达,CCK-8法检测细胞增殖能力,细胞划痕方法检测细胞迁移能力,Transwell小室法检测细胞侵袭能力,Western blot法检测ER-α受体蛋白表达水平。结果:转染miR-107 inhibitor的Ishikawa细胞中miR-107表达水平显著降低,细胞的增殖迁移和侵袭能力降低,细胞中ER-a受体蛋白表达水平增高。结论:下调miR-107表达可能通过上调ER-α表达抑制Ishikawa细胞的增殖迁移和侵袭能力。
